摘  要:
Mitochondria are key organelles supporting energy homeostasis and signal transduction within cells, which are dynamically controlled by structure dynamics of the organelle network. We observed two different potential aspects of membrane surface condensates that is related to mitochondrial membrane structure. On the outer mitochondrial membrane, surface condensates of mitochondrial fission factor (Mff) regulate the fission machinery organized by dynamin-related protein 1 (Drp1). Phase demixing between Mff and Drp1 generates a membrane tension gradient that modulates fission activity. In the mitochondrial matrix, mitochondrial transcription factor A (TFAM) senses the curvature of the inner mitochondrial membrane, and this curvature sensing contributes to mtDNA organization. Together, these findings reveal a general mechanism by which mitochondrial membranes integrate structural and biochemical cues across distinct submitochondrial compartments. By coordinating fission at the outer membrane with genome organization near inner membrane, curvature-sensitive surface condensates may help mitochondria adapt their morphology, bioenergetic state, and gene expression to changing cellular demands. This framework provides a physical basis for understanding how mitochondrial architecture is coupled to organelle function and may offer new insights into diseases associated with disrupted mitochondrial dynamics and mtDNA homeostasis, and for how membrane and condensates coordinates within cells at subcellular level.
