Selective transport among organelles and the plasma membrane is fundamental to many cellular processes including control of protein and lipid levels, and cell signaling. Coat protein complexes (CPCs) select certain membrane-bound cargoes, package the cargoes into vesicles and tubules, and deliver them to specific organelles. The retromer is a conserved CPC that controls the transport of a wide range of endosomal cargoes to trans-Golgi networks (TGN) or plasma membrane. In contrast to the best-studied CPCs (Clathrin, COPI, and COPII), the molecular mechanisms regulating the function of retromer are very poorly understood. My postdoctoral research has identified have discovered a novel actin regulatory complex, the five-component WASH complex, that regulates the maturation of the retromer coat (Jia et al, PNAS, 2010; Jia et al, MBoC, 2012), and a new component for the retromer coat (Jia et al, eLife, submitted). I will discuss how these discoveries advance our understanding of the retromer biology and related diseases such as Parkinson’s disease and Alzheimer’s disease.
