The Klein laboratory studies mechanisms of Wnt signaling in early development and in the control of hematopoietic stem cell (HSC) self-renewal decisions. Wnt signaling is essential for establishing dorsal-ventral pattern in the embryo and also plays a central role in maintaining the capacity for self-renewal in diverse stem cell populations, including HSCs, mouse embryonic stem cells, epidermal, neural, and intestinal stem cells. In this seminar, I will present a mechanism to explain how Wnts inhibit GSK-3 and how this leads to the regulation of both ß-catenin and mTOR. This mechanism provides insights into the regulation of HSC homeostasis and has led to a method to maintain the self-renewing capacity of HSCs ex vivo. I will also discuss a novel mechanism by which embryos remember a transient Wnt signal and respond at a later stage by encoding a memory of the signal within the chromatin of developmental control genes.
