Recent advance in sequencing techniques has generated massive amount of data for the epigenome of a variety of cell lines and tissues. In contrast to data generation, data analysis has become the most rate-limiting step for scientists to gain biological insights from these high-throughput data. My research focuses on the characterization of the global epigenetic regulatory networks during cellular development, differentiations and pathology by using systems biology approaches, in particular data integration. In this seminar, I will introduce the application of data integration of deep sequencing data to address several biological questions including targeting of chromatin remodeling complexes (Genome Research 2011), regulation of ESC self-renewal and differentiation by the histone variant H2A.Z (Cell Stem Cell 2013), gene regulation by the transcription factor c-Myc (Cell 2012), and a catalog of long intergenic noncoding RNAs in T cells (Nature Immunology 2013). In future, I am planning to investigate the epigenetic regulation of long noncoding RNAs and their functions in maintaining cell identity and in directing cellular differentiation in immune cells.
