C-reactive protein (CRP) and Serum Amyloid A (SAA), have long been used as clinical markers to monitor the progress of infection and evaluate the outcome of therapeutic treatments. However, the exact physiological functions of CRP and SAA still remain largely unknown. We aim to understand the structural and functional aspects of the interaction between CRP/SAA and their cell surface receptors, particularly in human autoimmune diseases and chronic inflammatory diseases. So far, 1) we have established the structural recognition and functional activation of antibody IgG and IgA Fc receptors and its homolog Serum Amyloid P (SAP). 2) We have determined the first crystal structure of SAA, demonstrating that the loss of the structural integrity of SAA would be critical to the amyloid formation. Taken together, our mechanistic studies shed new light on the understanding of CRP and SAA proteins` functions in the immune systems and will provide therapeutic guiding significance for the treatment of important human diseases.
