The structural, functional and dynamic complexity of the brain presents great challenges to neuroscientists. A large part of the structural complexity of the brain lies in its cellular heterogeneity. Diverse celltypes, as the building blocks of neural circuits, are highlyi ntermingled and interconnected but display distinct physiological properties, connectivity patterns and gene expression profiles.One importants towards understanding the inner working of the brain isthedissection of neural circuit assembly by examining the mechanisms underlying the specification, maturation and plasticity of individual celltypes, as well as their dysfunction under pathological conditions. Utilizing genetic approaches engaging the intrinsic gene regulatory mechanisms that generate, maintain or correlate with celltype identity and phenotypes, we established experimental systems which promise precise and reliable identification and manipulation of distinct GABAergic celltypes in mouse neocortex. miRNA expression profiling in these celltypes using novel microRNA tagging and affinity-purification(miRAP) strategy revealed extensive differentially expression in glutamatergic and GABAergic neurons and subtypes of GABAergic neurons. Future work centering on mouse genetic engineering and incorporating imaging, electrophysiology, genomics/epigenomics, and behavior analysis, will lead to a better understanding of the organization, development, and function of the cerebral cortex and help achieving a biological explanation of mental disorders.
