DNA replication is a fundamental process whose deregulation can challenge genome stability and lead to mutations, cancer and other genetic diseases. However, in eukaryotes and especially in metazoa, the mechanisms that control the replication program, i.e. where and when replication initiates, remain largely unknown. We developed a novel method that allowed us to generate one of the first high-resolution human replication timing profiles by massive sequencing. Studies of replication timing profiles of various cell types revealed that the genome is organized in megabase replication domains. Analysis of chromatin properties and 4C experiments showed that these replication domains correspond to higher-order chromatin structural units. Taking advantage of these data we further demonstrated that replication plays an important role in shaping gene organization and mutation patterns during human genome evolution.
