Title: Notch signal pathway——an ancient cell interaction mechanism
Abstract:
From the very beginning, Notch gene was found by Morgan and his coagent, because the partial dysfunctiong of this gene cause notch wing. However, its fundamental role in differentiation, proliferation, and apoptotic programs was found gradually. now, we have known that the notch signal pathway is one of the most important and conserved pathway in metazoan development.
Notch signal pathway rely on cell-to-cell interaction, both of the Notch receptor and its ligands, including Delta and Serrate (in mammals, Delta-like and Jagged) are transmembrane proteins. ligand-receptor binding promotes two proteolytic cleavage events in the Notch receptor, which produce NECD (Notch extracellular domain) and NICD(Notch intracellular domain). NECD is endocytosed by the ligand-expressing cell, and NICD moves to nucleus of the receptor-expressing cell and regulate gene expression. The downstream effect of both of them are hot and important topic. Another important topic is how cells regulate the Notch signal pathway by regulating the output, strength and duration of signal. Recent study show that even the cell-cell contact area can affect Notch signaling.
